Preparing Plating Bacteria

Use a streaked plate of su- 7155 bacteria not more than 1 month old and has been stored in the cold room.

Make an overnight culture of 1 colony in 25 ml of LB media in a 125 ml sterile, baffled flask. Shake in 37 degree incubater overnight.

In the morning, transfer 3 ml of the overnight culture into 300 ml of LB media in a 1 liter sterile, baffled flask.
Shake for 5 hours at 37 degrees.

Pour into sterile GSA centrifuge tubes, balance, spin 5K, 10 minutes. Resuspend gently with pipet, in 25 ml fresh LB media.

Transfer to sterile dropper bottle.

Use for plating phage, up to 2 weeks, but better for only 1 week.
