Using the Densitometer (in Huber Lab)

1) Using: Molecular Dynamics, Computing Densitometer, ImageQuant software

2) Turn on 2 toggles on side of densitometer and computer front switch. Warm up 15 min.

3) Click on the ImageQuant icon to start program.

4) Under…Scanner, select Calibrate. Takes 5 minutes, verrrrrrry slow. Do not proceed until complete.

5) Place gel on glass plate, leaving room on all sides to seal down with our saran wrap. Dry the plate well, then seal down the saran to the plate, should seal well without tape and bunching. Close lid.

6) Under…Scanner, select sample, give coordinates of area ie. D4 to H11, name id for sample, do not press enter until everything is correct. Enter = Scan

7) You should see your sample on the screen.

Notes: if your bands are not very clear, the instrument did not take a good picture. You can try to let it warm up 2 hours and try it again for weak bands. Not much control.

8) On the Dell computer near the system, Click: Start, Programs, WF-FTP from the bottom left button. Connect to densitometer as anonymous. Select the file and send over to your AFS space (using the H drive selected on the left, press “<=”). Files transfer as binary.

9) Close screen. Open IQ, adjust image by using the histogram adjustment.

10) Save as…TIF file. Be sure the file name ends with .TIF then press enter.
